Anti-TTF-1/NKX2.1
“Thyroid Transcription Factor-1 (TTF-1), also known as NKX2.1, is a homeodomain-containing transcription factor that plays an essential role in the embryonic development, differentiation, and functional maintenance of the thyroid gland, lung, and ventral forebrain. NKX2.1 regulates the expression of several tissue-specific genes, including thyroglobulin, thyroid peroxidase, surfactant proteins, and Clara cell secretory proteins, thereby contributing to normal thyroid and pulmonary function. In adult tissues, TTF-1 expression is primarily restricted to thyroid follicular epithelial cells and respiratory epithelial cells, particularly type II pneumocytes and club cells.
TTF-1 is one of the most widely utilized lineage-specific markers in diagnostic surgical pathology. Its expression is frequently retained in primary lung adenocarcinomas, small cell carcinomas of the lung, and tumors of thyroid origin, making it highly valuable in determining the primary site of metastatic carcinomas. Loss or altered expression may occur during tumor progression and dedifferentiation in certain malignancies.
