DNA topoisomerase II alpha (Topo-II?) is an essential to maintain the topology of DNA, it up-regulated in different tumors. Topo II is maintains chromatin condensation and separation during mitosis. DNA topoisomerase II alpha levels are cell cycle regulated and the expression level increases between G2 to M phase after replication phase. DNA topoisomerase II alpha has been linked with maintenance of DNA integrity during ?cell proliferation and it may be the main isoform of Topo II involved mitotic processes. Topo II ? passes one strand of DNA through a reversible break in a second DNA strand, which catalyzes the topological isomerization of DNA during cell cycle. Due to its significance, Topo II ? overexpression is the target for many chemotherapeutic agents. The majority of anticancer drugs targeting Topo II? initiate apoptosis by stabilizing the reversible covalent complex formed between DNA and Topo II?
