Serum Amyloid P (SAP), a member of pentraxin family, is a glycoprotein which has a characteristic pentameric organization. It is an acute phase protein, structurally related to C-reactive protein. SAP is made by hepatocytes and secreted into the blood. SAP is the single normal circulating protein that shows specific calcium-dependent binding to DNA and chromatin in physiological conditions. The avid binding of SAP displaces H1-type histones and thereby solubilizes native long chromatin, which is otherwise profoundly insoluble at the physiological ionic strength of extracellular fluids. The normal plasma protein serum amyloid P component (SAP) binds to fibrils in all types of amyloid deposits, and contributes to the pathogenesis of amyloidoses.
