“SALL4 is a zinc-finger transcription factor that plays a critical role in embryonic development, stem cell pluripotency, self-renewal, and lineage commitment. During normal human development, SALL4 is highly expressed in embryonic stem cells and germ cells, while expression is largely absent in most differentiated adult tissues. Aberrant re-expression of SALL4 has been reported in a variety of malignancies, including acute myeloid leukemia, myelodysplastic syndrome, hepatocellular carcinoma, germ cell tumors, gastric carcinoma, and other aggressive solid tumors. SALL4 contributes to oncogenesis through regulation of stemness-associated pathways, epigenetic remodeling, cell proliferation, and inhibition of differentiation.
Loss or gain of SALL4 expression has significant biological and clinical implications. In cancer, elevated SALL4 expression is frequently associated with poor prognosis, increased metastatic potential, treatment resistance, and maintenance of cancer stem-cell phenotypes. Consequently, SALL4 has emerged as a diagnostic biomarker and a potential therapeutic target in multiple tumor types.
