The p21/WAF1 a cyclin-dependent kinase inhibitor protein encoded by p21/WAF1 gene and and one of the key p53 regulated gene product and it mediates cell cycle growth arrest in the absence of p53 expression and also plays a significant role of a tumor suppressor in cancer. It induces cell growth arrest due to cyclin dependent kinase inhibition activity, and maintains G2 cell cycle arrest by inhibiting the interaction of PCNA with Cdc25C and other activated protein kinases. Recent studies show that p21 may possibly act as an oncogene, because it inhibits apoptosis in the absence of p53 and induce cell proliferation in some tumors. These studies bring out the “antagonistic duality” of p21, since it shows both the pro oncogenic and anticancer properties at the same time(2).
