MutS homologue 1 (MSH1) is a member of Mutator S (MutS) DNA mismatch repair protein family. MSH1 binds to DNA mismatches to maintain the integrity of genetic information, initiate DNA repair by forming heterodimer complexes with MSH2, MSH6 and PMS2. Strand misalignment can occur during DNA replication, resulting in microsatellirte repeats, referred to as microsattelite instability (MSI). These Strand misalignments in DNA repair pathways are associated with human carcinogenesis. MLH1 gene Mutations results in increased incidence of tumors with MSI, such as in some forms of colon cancer e.g. Hereditary Non Polyposis Colon Cancer (HNPCC), a subset of sporadic carcinomas and breast cancer. Loss of expression of MLH1 has also been reported in acute lymphoblastic leukemia, endometrial carcinoma, Gastric carcinoma and ovarian carcinoma
