Both IHC (Immunohistochemistry) and IF (Immunofluorescence) have same functions. However, IF is the staining method usually used for multiplexing, particularly when co-localization is required or when 2 or more than 2 different antibodies needs to be detected on a single tissue.

IF follows the protocol same like IHC with slight modifications. The basic differences between IHC and IF is the use of fluorescent tagged (conjugated) antibodies in IF which may have colors like FITC, PE, Cy3, Cy5, Texas Red etc. are attached either with primary antibodies or secondary antibodies. After that there is no chromogen required here and final mounting with DAPI follows. For visualization, fluorescent microscope is also required.

ISH- ISH has broad applications such as physical chromosome mapping, karyotyping, and pathogen profiling. Riboprobe also enables the localization and analysis of gene expression profile.

FISH- As a diagnostic tool, FISH is widely used in clinical genetics, neuroscience, reproductive medicine, toxicology, microbial ecology, evolutionary biology, comparative genomics, cellular genomics and chromosome biology to check for genetic abnormalities associated with prenatal diagnosis, cancer, birth defects and mental retardation. It is possible to differentiate benign and malignant tumors in the same tissue using FISH.

There are two basic approaches to visualize the DNA or RNA target sequences in situ: chromogenic in situ hybridization (CISH) and fluorescence in situ hybridization (FISH). While these approaches involve the same underlying principle, the instrumentation required to visualize the specimen is different.


The nucleic acids within a histologic specimen can be detected by the hybridization of a complementary nucleic acid probe in which a reporter molecule is attached. Common non-radioactive labels for probe are fluorescein and digoxigenin. Subsequent visualization of the reporter molecule localizes DNA or RNA sequences in tissue or cells samples.


In Situ Hybridization (ISH) is a powerful technique for precise detection and localization of a specific nucleic acid sequence within a histologic section.

Graphical User Interface for Xmatrx is based on our popular i6000 interface. Xmatrx GUI expands the i6000 interface to enrich your programming experience. Main interface window displays the slide map and control panel. Slide map offers an intuitive and convenient way of selecting & editing slide protocols and to query the progress of a slide during the run.

Various operations are color-coded. Slide objects on the main window are displayed in appropriate colors based on their current operation making it convenient to get a snapshot of the run status from a distance. In the event of an error condition, system plays out an appropriate audio message and also visual message on the control panel.


Need for updates/bug fixes, if any, determines the frequency of new software releases.  There could be 1-2 releases every 10 – 12 months.

No. System discards the tips and Coverslips after use. Reuse of these discarded tips and Coverslips is not recommended, as it will cause cross contamination and also damages the instrument functionality.


While applying large volumes (more than 100ul), Xmatrx system uses large tips. Typically, volume range for this step is 100-850ul. 


While processing expensive reagents like AR Solution, Antibody/Probe, Xmatrx system uses small tips. Typically, volume range for this step is 10-100ul.

A reagent vial is entered into the list either or when the system reads the Barcoded vial for the first time. If the reagent does not exist in the database, the vial will have to be returned back to BioGenex, to be replaced by a new vial. This allows the system to track reagent usage, lot numbers and expiry to ensure that the correct reagent is being used for Xmatrx Elite only.

In case, if any antibody is not available fromBioGenex, the User will have an option to buy an empty Barcoded vial to run their own Antibody for Elite. This includes just a basic cleaning of the instrument to prevent any contamination and to ensure proper functionality of the instrument. We recommend basic cleaning done by the user as per the Maintenance Chart.


Xmatrx Elite / Infinity is provided with a standalone systems. So the user will not require any extra bench space. The unit will be received as a cabinet with all the bulk reagents placed within it.


No the instrument does not need to be under a hood.


No you do not need to calibrate the instrument every time you use it. The instrument will be calibrated when it is installed. Re-calibration will be required if the instrument is moved to a different location.


BioGenex field service engineer will install the instrument. Your local sales repsentative and application scientist will be involved in training and optimizing your protocols on the instrument.


FFPE tissues, pap smears and cell smudges can be used with these probes.


We provide a universal detection kit for all probes (Super SensitiveTM One-Step ISH detection kit).


BioGenex probes are intended to use with Super SensitiveTM One Step ISH detection kit and optimized for chromogenic visualization under bright field microscopy. However, if the end-user wants to use any other kit and visualization technique, they need to validate their own protocol.


For the less no. of slides, if you are using BioGenex antigen retrieval solutions {HK546 (EZ-AR™ 1 Elegance ), HK547 (EZ-AR™ 2 Elegance)}, you can reuse these solutions up to 2-3 times.


Following are the solutions for doing Antigen Retrieval without EZ-RT System and Microwave-

  1. Water bath- Set the temp. of water bath at 100’C. Take one glass/plastic container and pour AR solution in it. Thereafter put the slides (with slide holder) in glass/ plastic container. After that put this container (having slides) into water bath for 30 mins. Make sure that the water bath water should not mix with the AR solution container. Heat at 100’C for 30-40 min and take out the container from water bath. Thereafter follow the regular IHC protocol.
  2. Hot plate- Set the temp. 100’C. Take AR solution in glass container and then put slides into glass container. Put that glass container at hot plate and heat for 30-40 min at 100’C. Thereafter follow the regular IHC protocol.

Note- Count the time of AR once AR solution temp reaches to 100’C. If it is not possible to reach to 100’C then it can be done at 98’C also.

Most antibodies can be used either with DAB or AEC chromogen depending on the preference for staining under a light microscope.


The requirement of antigen retrieval depends on the antibody used and its staining protocol. For frozen and de-calcified tissues, heat-based antigen retrieval is NOT recommended. We provide alternate solutions for these kinds of tissues such as the Decal Retrieval Solution for acid-decalcified bone marrow tissue and enzymes for tissue digestion.


The proprietary technology used in the EZ-AR Elegance solutions provides stronger intensity and reproducible results. These solutions are designed for used with automated platforms such as Xmatrx systems and the EZ Retriever. The normal antigen solutions are intended for use with manual protocols.


Evaporation in Citra and Citra plus antigen retrieval solution is more and its ph also changes after use that’s why these can not be reused again and again.


For concentrated antibodies, the protein concentration is available on the data sheet for that specific antibody. Ready-to-Use (RTU) antibodies are pre-diluted for optimal staining results. We can provide the concentration of RTU antibodies upon request.


There are various reasons as to why we discontinue certain antibodies ranging from raw material issues to insufficient sales. The replacement products, if available, can be provided on request.


Most primary antibodies raised in mouse (IgG +IgM) and rabbit (IgG) work with our Poly-HRP Secondary antibody. In case of primary antibodies raised in other species, we also provide species-specific secondary antibodies for use with our Link-Label System.


The one-step detection kit is designed with proprietary technology which eliminates the use of Super Enhancer required by the two-step detection kit. Therefore, the one-step kit provides superior sensitivity and specificity with the advantage of a short protocol.

BioGenex antibodies are validated and intended for use on human FFPE and frozen tissues. If used on tissues from other species, the protocol must be validated by the end user.


While using the EZ-Retriever®System for antigen retrieval, you have the facility to increase and decrease the temp and time according to your need. You can use 107 Deg C maximum temperature and 15 mins maximum time in one cycle with BioGenex antigen retrieval solutions {HK521 (EZ-AR™ 1), HK546 (EZ-AR™ 1 Elegance), HK522(EZ-AR™ 2), HK547 (EZ-AR™ 2 Elegance)}. If you are using other solutions then you can use it at 95 Deg C for 10 min in one cycle.


BioGenex provides information on the specific antigen retrieval solution recommended for use with its primary antibodies. We offer Retrievit Sampler Kit for use with primary antibodies from other manufacturers. This kit provides the customer with five different antigen retrieval solutions so the user can determine which one is the best for that particular antibody.


The microwave pressure cooker method is a procedure that utilizes a microwave with a pressure cooker for antigen retrieval and staining. The pressure cooker sold by BioGenex is especially designed for use in a microwave oven. Its advantages include the speed of treatment, reproducibility of results with large batches of slides, ability to use metal slide racks, and economy of equipment costs.


STAT (Latin: statum, meaning: to rush, urgent) Access allows the addition of emergency specimen slides for staining during an ongoing run. STAT slides will be processed and completed at the highest priority. During the processing of STAT slides, all other slides being processed shall remain on hold and are buffered regularly to keep the tissue hydrated. After completion of the STAT slides, all pending slides that are on hold will be processed according to their protocol.


Most autostainers on the market do not allow for the addition of new samples for staining when a run is in progress. Xmatrx Diagnostics breaks this limitation by enabling continuous operation of the system. The user can add new slides in the middle of a run if empty slide positions are available. The slides that are loaded in continuous mode will be processed along with the current slides without affecting the current run. Continuous Access enables the user to remove completed slides and load new ones without the need to wait for run completion. In addition, reagents that are no longer required can be removed and new ones can be added.

Graphical User Interface for Xmatrx is based on our popular i6000 interface. Xmatrx GUI expands the i6000 interface to enrich your programming experience. For the less no. of slides, if you are using BioGenex antigen retrieval solutions {HK546 (EZ-AR™ 1 Elegance ), HK547 (EZ-AR™ 2 Elegance)}, you can reuse these solutions up to 2-3 times.


Following are the solutions for doing Antigen Retrieval without EZ-RT System and Microwave-

  1. Water bath- Set the temp. of water bath at 100’C. Take one glass/plastic container and pour AR solution in it. Thereafter put the slides (with slide holder) in glass/ plastic container. After that put this container (having slides) into water bath for 30 mins. Make sure that the water bath water should not mix with the AR solution container. Heat at 100’C for 30-40 min and take out the container from water bath. Thereafter follow the regular IHC protocol.
  2. Hot plate- Set the temp. 100’C. Take AR solution in glass container and then put slides into glass container. Put that glass container at hot plate and heat for 30-40 min at 100’C. Thereafter follow the regular IHC protocol.

Note- Count the time of AR once AR solution temp reaches to 100’C. If it is not possible to reach to 100’C then it can be done at 98’C also.

Most antibodies can be used either with DAB or AEC chromogen depending on the preference for staining under a light microscope.


The requirement of antigen retrieval depends on the antibody used and its staining protocol. For frozen and de-calcified tissues, heat-based antigen retrieval is NOT recommended. We provide alternate solutions for these kinds of tissues such as the Decal Retrieval Solution for acid-decalcified bone marrow tissue and enzymes for tissue digestion.


The proprietary technology used in the EZ-AR Elegance solutions provides stronger intensity and reproducible results. These solutions are designed for used with automated platforms such as Xmatrx systems and the EZ Retriever. The normal antigen solutions are intended for use with manual protocols.


Evaporation in Citra and Citra plus antigen retrieval solution is more and its ph also changes after use that’s why these can not be reused again and again.


For concentrated antibodies, the protein concentration is available on the data sheet for that specific antibody. Ready-to-Use (RTU) antibodies are pre-diluted for optimal staining results. We can provide the concentration of RTU antibodies upon request.


There are various reasons as to why we discontinue certain antibodies ranging from raw material issues to insufficient sales. The replacement products, if available, can be provided on request.


Most primary antibodies raised in mouse (IgG +IgM) and rabbit (IgG) work with our Poly-HRP Secondary antibody. In case of primary antibodies raised in other species, we also provide species-specific secondary antibodies for use with our Link-Label System.

While using the EZ-Retriever®System for antigen retrieval, you have the facility to increase and decrease the temp and time according to your need. You can use 107 Deg C maximum temperature and 15 mins maximum time in one cycle with BioGenex antigen retrieval solutions {HK521 (EZ-AR™ 1), HK546 (EZ-AR™ 1 Elegance), HK522(EZ-AR™ 2), HK547 (EZ-AR™ 2 Elegance)}. If you are using other solutions then you can use it at 95 Deg C for 10 min in one cycle.


BioGenex provides information on the specific antigen retrieval solution recommended for use with its primary antibodies. We offer Retrievit Sampler Kit for use with primary antibodies from other manufacturers. This kit provides the customer with five different antigen retrieval solutions so the user can determine which one is the best for that particular antibody.


The microwave pressure cooker method is a procedure that utilizes a microwave with a pressure cooker for antigen retrieval and staining. The pressure cooker sold by BioGenex is especially designed for use in a microwave oven. Its advantages include the speed of treatment, reproducibility of results with large batches of slides, ability to use metal slide racks, and economy of equipment costs.


STAT (Latin: statum, meaning: to rush, urgent) Access allows the addition of emergency specimen slides for staining during an ongoing run. STAT slides will be processed and completed at the highest priority. During the processing of STAT slides, all other slides being processed shall remain on hold and are buffered regularly to keep the tissue hydrated. After completion of the STAT slides, all pending slides that are on hold will be processed according to their protocol.


Most autostainers on the market do not allow for the addition of new samples for staining when a run is in progress. Xmatrx Diagnostics breaks this limitation by enabling continuous operation of the system. The user can add new slides in the middle of a run if empty slide positions are available. The slides that are loaded in continuous mode will be processed along with the current slides without affecting the current run. Continuous Access enables the user to remove completed slides and load new ones without the need to wait for run completion. In addition, reagents that are no longer required can be removed and new ones can be added.
